Biological Evaluation and Conformational Preferences of Ferrocene Dipeptides with Hydrophobic Amino Acids

چکیده
Despite the large number of peptidomimetics with incorporated heteroannularly functionalized ferrocenes, few studies have investigated their bioactivity. Here, we report biological evaluation and conformational analysis enantiomeric dipeptides derived from 1′-aminoferrocene-1-carboxylic acid (Fca) hydrophobic amino acids (AA = Val, Leu, Phe). The properties Y-AA-Fca-OMe (Y Ac, Boc) were elucidated by experimental (IR, NMR, CD, X-ray) theoretical (DFT) methods. prepared screened for antimicrobial activity against selected Gram-positive Gram-negative bacteria, lactic bacteria yeasts, while antioxidant was tested DPPH FRAP Of all compounds tested, dipeptide d-2a showed best antibacterial S. aureus, B. subtilis, P. aeruginosa at a concentration 2 mM. time–kill curves that concentration- time-dependent. Chirality (d-) more polar-protecting group (Ac) found to affect activity, both antioxidant. All peptides are considered be highly chemically stable in acidic buffer media. Dipeptides d-1a–3a, which subjected determination proteolytic revealing very good resistance proteolysis presence chymotrypsin.

Structure-based conformational preferences of amino acids.
Proteins can be very tolerant to amino acid substitution, even within their core. Understanding the factors responsible for this behavior is of critical importance for protein engineering and design. Mutations in proteins have been quantified in terms of the changes in stability they induce. For example, guest residues in specific secondary structures have been used as probes of conformational ...
متن کاملInteraction of human interferons with immobilized hydrophobic amino acids and dipeptides.
Human fibroblast interferon binds to L-tryptophan, D-tryptophan, L-phenylalanine, and L-tyrosine, all immobilized directly to cyanogen bromide-activated agarose, as well as to L-tryptophan and D-tryptophan methyl ester, both immobilized via molecular arms. The retention of fibroblast interferon is selective and results in a 2300-fold purification. Human leukocyte interferon binds neither to L-t...
متن کاملConformational preferences of amino acids in globular proteins.
In a previous paper [Levitt, M., and Greer, J. (1977), J. Mol. Biol. 114, 181--239], an objective compilation of the secondary-structure regions in more than 50 different globular proteins was produced automatically. In the present paper, these assignments of secondary structure are analyzed to give the frequency of occurrence of the 20 naturally occurring amino acids in alpha helix, beta sheet...
متن کاملThe conjugates of ferrocene-1,1'-diamine and amino acids. A novel synthetic approach and conformational analysis.
A novel synthetic approach toward a poorly explored bioorganometallic consisting of ferrocene-1,1'-diamine bearing structurally and chirally diverse amino acid sequences is reported. Until now, ferrocene-1,1'-diamine was suitable for accommodating only identical amino acid sequences at its N-termini, leading to the symmetrically disubstituted homochiral products stabilized through a 14-membered...
